Using U¹³C₆-cystine infusion, we show tail vein achieves robust steady-state labeling of cystine and downstream metabolites (glutathione, taurine pathways) without the supraphysiologic tracer accumulation seen with jugular infusion.

Our solution is a tail vein catheter that can be implanted in just 5-10 min under brief anesthesia (vs 30-40 min jugular surgery). We secure the catheter and thread the tubing subcutaneously into a harness system. Mice can be infused awake. No surgery, no recovery, no analgesics needed!

YES! Just 2 hours of isoflurane profoundly altered metabolism. Glycolysis/TCA ↓, sulfur amino acids ↓, acylcarnitines ↓, while BCAAs ↑, ketones ↑, fatty acids ↑—suggesting complex metabolic deregulation. These pool size changes compromise flux interpretation.
This project started when our jugular surgeon left research. After initial panic, we wondered “can we use the tail vein?” But we would need anesthesia, is it really that bad for metabolism?
