Really depends what you're looking for. For us we aim for 1 mutaton/ORF and only need to do one round to screen 3-5K colonies - this approaches saturation for an avg size bacterial transcription factor. Get a variety of GOF and LOF, including a few rare.
We use mutazyme II for ORF, assemble into vector or double-cross over product by Gibson, and couple to reporter assay (to detect increase/decrease in expression). Of course bacteria, so easy

TLDR: both studies used experimental evolution comparing single & multiple species in biofilms.
They show strong selection to inhabit diverse biofilm niches that single species can do well w/ few mutations, but multiple species can do better, with only 1-2 muts!

I was sad to learn that my postdoctoral mentor, David Botstein, died yesterday. I started with David as a postdoc in 1998, and he had a profound effect on both my life and scientific career. He was a giant in the field of genetics, making seminal contributions in both yeast and human genetics. 1/
