Enhanced sensitivity and scalability with a Chip-Tip workflow enables deep single-cell proteomics - Nature Methods Chip-Tip is a label-free quantification-based single-cell proteomics workflow for deep single-cell proteomics, which identifies over 5,000 proteins and 40,000 peptides in single HeLa cells.

🚨 Our #singlecell article is out! We describe the Chip-Tip label-free SCP workflow quantifying >5000 proteins in single HeLa cells. With high scalability, increased depth and throughput we can now envision large-scale LFQ-SCP biomedical studies!

Finally, an experienced MS group performed a proper comparison of MS plasma proteomics vs. Olink! Kudos to the Coon lab! Cannot wait to read all the details!

Not sure of the gradient/flow you are using but at lower flow it can often help to go to a lower ID on the emitter. That's solved a few problems for us, but not as extreme as what you are seeing.

Hello #TeamMassSpec and BioID fans! We are searching for particular lots of Cytiva Sepharose-Streptavidin resin. Recent lots have been poorly compatible with MS. If you have anything to spare from lots 10326476, 10312459, 10269523 or 10296366 please get in contact!

Even just diluting the provided standards in your lysis buffer can be effective. I did that a few days ago with a 562 nm BCA assay... thought it was Thermo or Pierce, unsure now.

IonOpticks is my go-to for most things these days. And in terms of heater setup for Thermo instruments it's quite affordable.

Nope. It's on the bottom of the stack. It's a fairly compacted design.

There are for sure bugs to work out. Like how 1 in 10 FAIMS runs randomly don't record data.

Where are your issues lying? It's pretty easy to run it via FragPipe but most of the trouble with these kinds of experiments is in setting up controls etc.

Generally this is done via PaSER/Bruker Proteoscape which works quite seamlessly via the HyStar queue. The PaSER diaPASEF search is based on an earlier version of DIA-NN making them quite comparable.
