💫The study, led by CNAG, @cabd-upo-csic.bsky.social and the Duke University Medical Center, is a further step to understand non-coding regulatory regions and 3D hubs that coordinate gene expression, laying the foundation for more precise genome engineering and modification of gene expression

These findings show how the same gene can play distinct roles in different tissues, as illustrated in mammalian sex determination. They also provide a foundation to better understand a group of conditions, such as Differences of Sex Development (DSD)

Using the geostatistics-inspired method, researchers identified a novel regulatory region controlling Fgf9, a key gene downstream of Sry that activates Sox9 and triggers testis development, as well as new regulatory networks including the Meis1 and Meis2 genes

To decode this vital process, CNAG developed 𝘔𝘌𝘛𝘈𝘓𝘰𝘤𝘪, a powerful predictive method that brings together a comprehensive and unified view of multiple genomic sites that had remained hidden, identifying 3D regulatory hubs and mapping gene regulation across cell types
2️⃣ 𝘛𝘩𝘦 𝘧𝘪𝘳𝘴𝘵 𝘴𝘵𝘦𝘱
To provide new answers to these questions, researchers began by obtaining the tiny gonadal cell populations before and after sex determination. Surprisingly, even using Hi-C and ChIP-seq, only minimal 3D chromatin changes were observed
