First dynamic in vivo analysis of dystrophic MuSCs. Identifies niche dysfunctions and points to strategies to restore stem cell function in DMD.
Cross-grafting experiments uncouple MuSC behaviours: Migration is dictated by the myofibre niche, fate decisions are fibre-independent, and symmetric cell divisions are disrupted. Resolves some long-standing debates in the field.
mdx stem cell dysfunction is driven by p38/PI3K signalling, unlike healthy MuSCs, leading to precocious differentiation and loss of stemness.
