Great work Daniela!
We are very happy to see our study finally appear online @nature.com! This has been work of nearly 10 years in collaboration with the National Institute of Genome Medicine π²π½, the National Cancer Institute π²π½, the @sangerinstitute.bsky.social and others β¬οΈ

Rosalind's companion piece to our genetic interactions manuscript. GRAPE, to analyze synthetic lethality screens, and Synulator, to generate simulated synthetic lethal gold standards because none exist today.

Excited to share my first first-author preprint!
This work would not have been possible without the support and guidance of my supervisor Dr. Elena Kuzmin, as well as my lab mates (past and present) and our collaborators.
with a great team of collaborators @traver.bsky.social @guilbourque.bsky.social
4οΈβ£ Why are we excited about this is because this expands our understanding of how chromosome arm deletions are buffered in triple negative breast cancer which should enable us to develop targeted therapies.
3οΈβ£ Functional assays revealed that chr4p loss is associated with disrupted mitochondrial respiratory function and reduced glycolytic capacity, suggesting metabolic rewiring. Increased reactive oxygen species and lipid peroxidation compromised antioxidant defense mechanisms.
2οΈβ£ We analyzed DepMap pooled genome-wide CRISPRCas9 screens and identified complex genetic interactions specific to chr4p deletion in basal breast cancer cell models.
1οΈβ£ We used differential gene expression analysis of patient derived xenografts and cancer cell models to show that chr4p loss is associated with changes in cellular energetics.
Excited to share our preprint on the βIntegrated Metabolic Complex Genetic Interaction Network of Chromosome 4p Loss in Basal Breast Cancerβ led by @lynnkaram.bsky.social! www.biorxiv.org/content/10.1...
Thank you for highlight GSA!
In #G3journal, @elena-kuzmin.bsky.social et al. present PARPAL database, a valuable resource for #yeast researchers to understand the retention and evolution of paralogs. A phenomics screen further shows how proteins respond to paralog deletion by localization & abundance changes. buff.ly/oM97WyH
Our manuscript on the PARPAL database is out in G3! We share our phenomics screen on how proteins respond to their paralog deletion by localization & abundance changes and integrate other datasets with collab @guilbourque.bsky.social @leopoldparts.bsky.social doi.org/10.1093/g3jo...
At long last, Rosalind's paper on comparing methods for paralog synthetic lethality is online:
genomebiology.biomedcentral.com/articles/10....
8οΈβ£ Why are we excited about this is because this is the first systematic investigation of relocalization changes of paralogs and provides insight into their retention in evolution
7οΈβ£ Finally, our findings are consistent with non-canonical protein isoform abundance as a mechanism for paralog compensation
6οΈβ£ Redistributed proteins were encoded by functionally redundant paralogs and showed genetic and protein interactions in the subcellular compartments of their paralogs
